Antifungal susceptibility profile of Trichosporon isolates: correlation between CLSI and etest methodologies BJM
Lemes,Raquel M.L.; Lyon,Juliana P.; Moreira,Leonardo M.; Resende,Maria Aparecida de.
The aim of the present study was to evaluate the antifungal susceptibility profile of Trichosporon species isolated from different sources employing the Clinical and Laboratory Standards Institute (CLSI) method and E-test method. Thirty-four isolates of Trichosporon spp. and six CBS reference samples were tested for their susceptibility to Amphotericin B, 5-flucytosine, Fluconazole, Itraconazole, Voriconazole and Terbinafine. All species showed high Minimun Inhibitory Concentrations (MIC) for Itraconazole and susceptibility to Fluconazole, The comparison among the results obtained by the CLSI method and E-test revealed larger discrepancies among 5-flucytosine and Itraconazole. The present work provides epidemiological data that could influence therapeutic...

Antimicrobial effects of allicin and ketoconazole on trichophyton rubrum under in vitro condition BJM
Aala,Farzad; Yusuf,Umi Kalsom; Jamal,Farida; Rezaie,Sassan.
Dermatophytosis is caused by a group of pathogenic fungi namely, dermatophytes, is among the most prevalent infectious diseases worldwide. Azole drugs are widely used in the treatment of dermatomycosis, but can cause various side effects and drug resistance to the patients. Hence, for solving this problem can be used from the plant extract as alternative for chemical drugs. Allicin is a pure bioactive compound isolated from garlic was tested for its potential as a treatment of dermatomycosis in this study. This study evaluated the in vitro efficacy of pure allicin against ten isolates of Trichophyton rubrum and the MIC50 and MIC90 ranged from 0.78-12.5 µg/ml for allicin. The results revealed that the order of efficacy based on the MICs values, all isolates...

Candida glabrata among Candida spp. from environmental health practitioners of a Brazilian Hospital BJM
Savastano,Catarina; Silva,Elisa de Oliveira; Gonçalves,Lindyanne Lemos; Nery,Jéssica Maria; Silva,Naiara Chaves; Dias,Amanda Latercia Tranches.
Abstract The incidence of the species Candida albicans and non-albicans Candida was evaluated in a Brazilian Tertiary Hospital from the environment and health practitioners. In a 12-month period we had a total positivity of 19.65% of Candida spp. The most recurring non-albicans Candida species was C. glabrata (37.62%), generally considered a species of low virulence, but with a higher mortality rate than C. albicans. Subsequently, C. parapsilosis (25.74%) and C. tropicalis (16.86%) were the second and third most commonly isolated species. Considering the total samples collected from the emergency room and from the inpatient and the pediatric sector, 19.10% were positive for Candida spp., with the predominance of non-albicans Candida species (89.42%). The...

Drug resistance of yeasts isolated from oropharyngeal candidiasis in aids patients Rev. Microbiol.
Silva,Maria do Rosário Rodrigues; Paula,Claudete Rodrigues de; Silva,Soraya Cristina; Costa,Théo Rodrigues; Costa,Márcio Rodrigues.
Candida spp was isolated from 59 (68.60%) out of eighty six samples of oral mucosa of AIDS patients. The identification, based or the production of a germ tube and chlamydospores, and on the assimilation and fermentation of carbohydrates, revealed 52 strains (88.13%) of C. albicans, 4 (6.77%) of C. tropicalis and 3 (5.08%) of C. krusei. The susceptibility of these strains to amphotericin B, flucytosine, itraconazole, fluconazole and ketoconazole was determined using the agar dilution method. Comparing the minimum inhibitory concentration values found in the susceptibility test with the serum levels achieved by these drugs, only 8.47% and 5.08% of the yeasts strains proved to be resistant to amphotericin B and flucitosyne, respectively. A high frequency of...

Genetic diversity and antifungal susceptibility testing of Trichosporon asahii isolated of Intensive Care Units patients BJM
Silva,Rosana Bellan de Oliveira; Fusco-Almeida,Ana Marisa; Matsumoto,Marcelo Teruyuki; Baeza,Lilian Cristiane; Benaducci,Tatiane; Mendes-Giannini,Maria José Soares.
Trichosporon asahii is an opportunistic pathogen, associated with a high mortality rate in immunocompromised patients. In this study, ten isolates, recovered from oral cavity and urine of patients in Intensive Care Units (ICU) over six months, were identified by classical and molecular methods, typed by RAPD and tested in vitro for susceptibility to fluconazole, itraconazole, 5-flucytosine and amphotericin B. A total agreement between the identification of Trichosporon sp by PCR based on sequences of the Internal Transcribed Spacer Regions (ITS) and on the sequences of small-subunit (SSU) ribosomal DNA (rDNA) was found. Randomly amplified of polymorphic DNA (RAPD), with primers P6 and M13, was used to determine the genomic profiles. The dendogram analysis...

Genetic polymorphism of clinical and environmental strains of Pichia anomala Biol. Res.
REYES,EUGENIO; BARAHONA,SALVADOR; FISCHMAN,OLGA; NIKLITSCHEK,MAURICIO; BAEZA,MARCELO; CIFUENTES,VÍCTOR.
In this work 20 clinical and 3 environmental yeast isolates were characterized by classical morphological and physiological methods, as well as molecular methods based on PCR of the ITS1-5.8S rDNA-ITS2 region. The characteristic morphology and biochemical profiles observed in these samples correspond to those described for the Pichia genera, more specifically to P. anomala. The profiles of susceptibility to five antifungal drugs were determined by two broth dilution methods. The results obtained by both methods were comparable and showed that clinical isolates presented more resistance to azoles, amphotericin B, and 5-fluorocytosine, than environmental ones did. By amplification and sequencing of internal transcribed spacers (ITS1 and ITS2) and the...

Treatment of invasive fungal infections: stability of voriconazole infusion solutions in PVC bags BJID
Adams,Andréa I.H.; Morimoto,Lucia N.; Meneghini,Leonardo Z.; Bergold,Ana M..
Voriconazole is a novel broad-spectrum antifungal drug, employed in the treatment of invasive fungal infections, and represents an alternative to amphotericin B treatment. The manufacturer recommends that any unused reconstituted product should be stored at 2ºC to 8ºC, for no more than 24 h, but no recommendations about i.v. infusion solutions are given. Previous works have reported on the stability of voriconazole in polyolefin bags and just one in 5% dextrose polyvinyl chloride (PVC) bags, at a 4 mg.mL-1 concentration. In this work, the stability of voriconazole as an i.v. infusion solution in 0.9% sodium chloride and in 5% dextrose, in PVC bags, at 0.5 mg.mL-1, stored at 4 ºC and at room temperature, protected from light, was evaluated. These infusion...
